Hydrodynamics inside wells of extra-high 24 square deepwell plates
Extra-high 24 square deepwell plates (well volume: 25 ml) are applied when culture volumes of 2.5 to 3.5 ml (as can be maximally used for regular 24 square deepwell plates) do not suffice.
With extra-high 24 square deepwell plates, culture volumes up till 8 ml can be used, preferably at 300 rpm on a shaker with a 50 mm amplitude.
Oxygen-transfer rates between 40 and 60 mmol O2 l-1 h-1 (corresponding to kLa values of 200-300 h-1) are readily attainable.
For the (slowed-down) high-speed videos below we used hitplates from HJ- Bioanalytik
